Inducing angiogenesis

Limited growth capacity without a blood supply - need nutrients for cancer cell survival Normal functions of angiogenesis include: wound healing, embryonic development, female reproductive cycling. Cancer takes advantage of a normal process Involves signalling input from several pro-angiogenic growth factors. "Angiogenic switch" - moment at which a tumor begins to overexpress pro-angiogenic factors, such as vascular endothelial growth factor (VEGF) - Delivery of oxygen and nutrients - Production of growth factors that benefit tumor cells - Metastases can exit through new tumor vasculature into systemic circulation Two additional components play a role in tumor neovasculature: - Pericytes are supporting cells that have long been associated with normal tissue vasculature; however, recent studies reveal that pericyte coverage is also important for tumor angiogenesis - Bone marrow–derived cells, such as macrophages and neutrophils, recruited to lesions and may help initiate the angiogenic switch
